Rapid processing of renal glomeruli for electron microscopy.

چکیده
Recently we reported a new fine-needle aspiration biopsy method to obtain single glomeruli for electron microscopy in diagnostic and investigative pathology of the kidney. The small size and fragmentary nature of the specimen obtained by this technique necessitated a special device for collecting and processing the tissue for electron microscopy (Pasternack et al., 1978). A new and simpler device has now been constructed, which allows the method to be applied to routine laboratory work and clinical diagnostics. With this new device it is possible to carry out the whole process from fixation to polymerisation of the specimen. Having used conventional processing methods for electron microscopy with the new device, we were tempted to try out previously described rapid processing methods (Estes and Apicella, 1969; Bencosme and Tsutsumi, 1970; Hayatand Giaquinta, 1970; Johannessen, 1973; Rowden and Lewis, 1974) for two reasons. Firstly, the fine-needle aspirate is small enough to guarantee adequate penetration of fixatives, buffers, and embedding materials in a shorter time than is needed when conventional methods are used. Secondly, drainage of the processing fluids allows a thorough exchange of the processing medium. This is thought to render the

STAG ES OF DEVELOPMENT OF RENAL GLOMERULI IN THE NEWBORN RAT KIDNEY
Glomerular development of the kidney was studied in newborn rats by electron microscopy. Four different stages of glomerular development were defined: vesicle fonnation, S-shaped body stage, capillary loop fonnation, and glomerular maturation. In the fust stage, the mesenchymal cells form a spheroid mass. This is followed by the S-shaped body stage in which clefts appear in the mass. Afte...

Background & Aim: Evaluation and classification of many renal lesions particularly glomerular diseases are only on the basis of renal biopsy. The clinician's request from the histopathologist is a timely and accurate diagnosis of renal damage which in addition to light microscopy is dependent on the assessment of immune deposits in renal biopsy and verification of immune deposits in Electro...
متن کاملPrecipitating antigen-antibody systems are required for the formation of subepithelial electron-dense immune deposits in rat glomeruli
This study was conducted to determine whether multivalent, precipitating antigens are required for formation of subepithelial electron-dense immune deposits in glomeruli. 2-nitro-4-azidophenyl (NAP) was conjugated with variable density to human serum albumin (HSA) to yield nonprecipitating (NAP3.1 X HSA and NAP11.4 X HSA) and precipitating (NAP19.7 X HSA) antigens with antibodies to the hapten....

I. Tissue Sampling, Allocation, and Fixative A. Sampling 1. Sample Size An adequate sample size must be obtained for diagnosis. Needle gauge has dramatic impact on sample obtained. Eighteenor 19-gauge needles give very small, narrow samples and often may have inadequate representation of vessels. For focal lesions involving a small number of glomeruli, 25 glomeruli may be needed for LM examinat...
متن کاملSequential postmortem changes of glomeruli. Their detection by scanning electron microscopy.
Postmortem changes were studied by scanning electron microscopy (SEM) in renal tissue from five species of animals. Specimens were collected at intervals up to 24 hours after death. Six distinct morphological alterations were identified in the renal glomerulus, including capillary loop constriction, changes in microvillus morphologic features, podocyte surface blebs, swelling or fusion of pedic...
